永利娱场城官网版app廖志华教授团队在青蒿素代谢调控研究方面取得新进展
青蒿(Artemisia annua)在我国具有悠久的用药历史,屠呦呦先生在中医典籍《肘后备急方》受到启发从青蒿中提取获得了一种治疗疟疾的倍半萜化合物青蒿素,拯救了亿万人的生命,因而荣获2015年诺贝尔生理医学奖。青蒿中青蒿素的含量较低,仅为干重的0.01%-0.8%,提高青蒿素含量成为该行业共同追求的目标。开展青蒿素生物合成代谢调控研究,不仅可以解析青蒿素生物合成时空特异性的分子机制,而且可为培育青蒿素含量高的青蒿品种提供具有自主知识产权的基因资源。近日,我校永利娱场城官网版app廖志华教授团队在青蒿素生物合成代谢调控研究上取得新进展,其两项研究成果分别于5月14日和6月16日在《Journal of Experimental Botany》和《Industrial Crops and Products》在线发表。
廖志华教授团队采用多学科研究手段,从青蒿中分离鉴定了bHLH家族转录因子AabHLH112,该基因在青蒿分泌型腺毛体中高表达,且受4℃低温诱导,能够通过与AaERF1启动子结合,从而间接激活青蒿素生物合成基因表达,过表达AabHLH112能够大幅度提高青蒿素含量,该研究解析了4℃低温促进青蒿素生物合成的机制。相关论文“The cold-induced transcription factor bHLH112 promotes artemisinin biosynthesis indirectly via ERF1 in Artemisia annua” 于5月14日在《Journal of Experimental Botany》在线发表。植物学博士向礼恩为该论文的第一作者,廖志华教授为通讯作者。基于青蒿全基因组分析,发现了在进化上与拟南芥PIF3相关的青蒿bHLH转录因子AaPIF3,该基因表达与青蒿素合成部位具有相关性,能够激活青蒿素生物合成基因表达,转基因结果表明AaPIF3能够用于培育青蒿素高产的青蒿。相关论文“Overexpression of AaPIF3 promotes artemisinin production in Artemisia annua” 于6月16日在《 Industrial Crops and Products》在线发表。植物学博士生张巧卓为该论文的第一作者,廖志华教授和张芳源博士为通讯作者。上述研究得到国家自然科学基金和上海交通大学资助。
青蒿素生物合成代谢调控和种质创新是廖志华教授团队长期的重要研究方向之一,与上海交通大学唐克轩教授团队长期密切合作,开展青蒿功能基因组、青蒿素生物合成与代谢调控,并承接唐克轩教授团队创制的青蒿优质种质的繁育工作。近5年来,廖志华教授作为通讯作者/共同通讯作者在《Molecular Plant》等期刊发表了一系列研究论文,相关研究论文如下:
